摘要
目的观察胰岛素对分离培养的大鼠血管平滑肌细胞(vascularsmoothmusclecells,VSMCs)增殖及胶原蛋白合成的影响。方法采用大鼠的腹主动脉血管平滑肌细胞分离培养,应用3H-TdR和3H-脯氨酸掺入方法检测胰岛素对大鼠VSMCs干预后,VSMCs内DNA合成以及胶原蛋白合成。结果胰岛素可促进处于静止状态的大鼠VSMCsDNA及胶原蛋白的合成,并呈现明显的浓度依赖关系,在40nmol/L的浓度时DNA及胶原蛋白的合成达到高峰,DNA及胶原蛋白分别处于36h和48h时合成最为显著。结论胰岛素可明显促进培养的VSMCs增殖及胶原蛋白的合成。
Objective To understand the role of insulin in the cytobiology of blood vessels. Methods The vascular smooth muscle cells (VSMCs) from SD rat were cultured and respectively treated with 1, 10, 20, 30, 40 nmol/L insulin for 24 h or 30 nmol/L insulin for 12, 24, 36, 48 h, or with DMEM solution containing 10% FBS as blank control. DNA synthesis and collagen protein synthesis of rat VSMCs were detected by ^3H- TdR incorporation and ^3H-proline incorporation. Results Insulin could significantly promote DNA synthesis and collagen protein synthesis of quiescent rat VSMCs in a dose-dependent manner. DNA synthesis and collagen protein synthesis reached the peak when insulin concentration was 40 nmol/L, and showed significant difference at 36 h and 48 h with that of the blank control. Conclusion Insulin could promote proliferation and collagen protein synthesis of cultured rat VSMCs.
