实验性兔PVR玻璃体增殖膜的免疫组织化学研究

摘要 目的探讨激活淋巴细胞与实验性增殖性玻璃体视网膜病变(PVR)的关系。方法将兔实验性PVR玻璃体增殖膜进行甲醛固定,石蜡包埋,制作切片,采用鼠抗人单克隆抗体CD3、CD43、CD68、CD23、CD20进行免疫组织化学染色研究;以兔实验眼的对侧眼球后段组织作阴性对照。结果20只眼中18只眼发生了视网膜脱离。实验中见到抗CD3、CD43、CD68阳性的染色结果,细胞膜及细胞浆内出现棕黄色颗粒,而CD23、CD20呈阴性。结论T淋巴细胞和组织细胞处于激活状态,在实验性PVR的发生发展过程中存在免疫介导过程。 Objective To explore the relationship between activated lymph cell and proliferative vitreous retinopathy (PVR). Methods The experimental PVR vitreous-proliferative membrane was treated by formalin fixation and paraffin inbedding,and immunohistochemistry-stain was performed by mice-anti-human monoclonal antibody CD3, CD 43, CD68, CD23, CD20. Posterior segment from the same rabbits'eye-ball was treated as negative control. Results Eighteen eyes(20 eyes) occurred retina detachment. Staining results from the experiments showed CD3, CD43, CD68 positive, and CD23, CD20 negative. Buffy particles appeared in cytoplasmic membrane and cytoplasm. Conclusion T-lyph cell and histiocyte are in activated state, indicating that immunjo-mediate procedure is present during experimental PVR.
