摘要
采用反相离子对高效液相色谱法测定肾上腺素与肾上腺酮.流动相为甲醇:25mmol/L醋酸钠-醋酸缓冲液(15:85,V/V),庚烷磺酸钠(5mmol/L)为离子对试剂,流速1.0mL/min,检测波长233nm,柱温25℃.在肾上腺素与肾上腺酮浓度分别为36-1800mg/L,36~1350mg/L范围内,其质量浓度与峰面积呈现良好的线性关系(r=0.9998,0.9995).当信噪比(S/N)为3时,二者的最低检测浓度分别为1.4和2.8mg/L.该法灵敏度高,可用于肾上腺素与肾上腺酮的定量分析.
Adrenalin and corticosterone were determined by an ion - pair reversed - phase chromatography with a mobile phase composed of methanol and 25 mmol/L acetate buffer ( contained 5 mmol/L sodium octanesulfonate, pH 4.7 ) ( 15:85 V/V). The operation was performed at the flow rate of 1.0 mL/min, and UV detection wave - length was 233nm. The results showed that the peak area and concentration showed excellent correlation, when mass concentration of adrenalin and corticosterone were 36 - 1 800 mg/L and 36 - 1350 mg/L ( r = 0.999 8,0.999 5) respectively. The detection limits of both adrenalin and corticosterone were 1.4 and 2.8 mg/L, if the rate of signal/noise was 3. All these indicated the method is suitable to analyze the adrenalin and corticosterone in broad areas.
