摘要
根据蔷薇科植物番茄红素β环化酶(LYC)基因的保守区序列,设计1对PCR引物,以枇杷基因组DNA为模板,采用PCR方法扩增长约300 bp的DNA片段,克隆入pMD-18T载体,测得该基因片段长312 bp,编码102个氨基酸,属于开放阅读框的一部分,不含内含子。在GenBank中进行同源性检索表明该序列与苹果番茄红素β环化酶基因同源性为97%,由此推测这个基因属于枇杷番茄红素β环化酶基因。
A pair of primer, designed from conserved regions of lycopene β-cyclase gene of rose family, is used to amplify homologous gene by polymerase chain reaction (PCR). As a result, one fragment about 300 bp is obtained, and then cloned into pMD-18T vector and sequenced. The result shows that the 312 bp belongs to a part of ORF (Open Reading Frame) with the code of 102 amino acids. It is suggested that the sequence is 97% identical to the lycopene β-cyclase gene from apple. Therefore, it can be deduced as lycopene β-cyclase gene fragment of loquat.
