摘要
目的探讨R hob tb2基因在多种肿瘤细胞中的表达及意义。方法以胎儿脑组织及人胚肺细胞为正常组织或细胞对照,选用八种不同组织的肿瘤细胞,用逆转录-聚合酶链反应法检测其R hob tb2基因表达。结果R hob tb2基因在乳腺导管癌细胞株T-47D中表达缺失,在乳腺腺癌细胞株M CF-7、SK-BR-3,以及卵巢癌、宫颈癌、膀胱癌、胃癌细胞中均有表达。筛选出R hob tb2基因表达缺失的乳腺癌细胞系T-47D,为R hob tb2基因功能研究提供基础和理想模板。结论R hob tb2基因可能以组织特异性或细胞特异性方式发挥抑癌基因作用。
[Objective] To investigate the expression and meaning of Rhobtb2 in different tumor cells. [Methods] Gene expression of Rhobtb2 in eight kinds of tumor cells was detected by RT-PCR. Fetal brain or fetal lung cell line was assigned as normal cell control. [Results] Rhobtb2 gene expression was observed in cell lines MCF-TSK-BR-3 of breast carcinoma, ovarian cancer cervical cancer, bladder carcinoma and stomach carcinoma but not in breast duetal carcinoma cell linesT-47D. The experiment screened out T-47D cell that lacked gene expression of Rhobtb2 and provided a perfect model for the further gene functional analysis, [Conclusions ] Rhobtb2 may act as a tumor suppressor gene in tissue or cell specificity manner.
