摘要
目的探讨重组人白细胞介素11(rh IL-11)对巨核细胞白血病细胞株D am i内转录因子GATA-1和NF-E 2表达的调节作用。方法用rh IL-11(50 ng/m l)刺激D am i细胞1、2、4 h,W estern b lot和RT-PCR法分析rh IL-11诱导D am i细胞GATA-1、NF-E 2 mRNA表达水平。结果rh IL-11可诱导D am i细胞GATA-1、NF-E 2mRNA表达,1 h后作用下降。结论rh IL-11可能通过诱导GATA-1、NF-E 2表达调控巨核细胞生成。
[Objective] To investigate the regulated action of recombinant human interleukin 11(rhIL-11) on transcription factors GATA-1 and nuclear factor erythroid 2 (NF-E2) in human megakaryocytic leukemia cell line Dami. [Methods] RT-PCR and Western blot were used to detecte the expression level of GATA-1 and NF-E2 mRNA in Dami cells induced by rhIL-11(50 ng/ml) for 1 h, 2 h, 4 h. [Results] rhIL-11 was found to increase the expression of GATA-1 and NF-E2 mRNA in Dami cell, but the action was decreased quickely after 1 hour, [Conclusion] rhIL-11 may take part in the regulation of megakaryocyte through increasing the expression of GATA-1 and NF-E2.
