扶正增效方对肺腺癌细胞放射增敏的基因表达谱分析 被引量:3

摘要 目的:采用基因芯片研究扶正增效方对肺腺癌细胞放射增敏作用的具体靶点。方法:将10只裸鼠皮下接种PAa细胞,并随机分为单纯放射组(Ⅰ组,5只)、放射加增效方组(Ⅱ组,5只)。自接种之日起,Ⅱ组给予扶正增效方汤剂灌胃,Ⅰ组灌服等容积生理盐水。瘤体生长至直径约1cm时,给予瘤体单次β电子线照射。照射后23h处死裸鼠,快速取瘤体,提取组织总RNA,分离mRNA。采用AffymetrixHumanGenomeU1332.0Array基因芯片分析两组之间基因表达谱的差异。结果:两组的基因表达谱显示,差异基因共有857条,上调349条,下调508条,其中39条在GeneBank中登录;按照功能分为细胞周期、凋亡、信号转导、代谢等类。结论:扶正增效方对肺腺癌细胞的放射增敏作用是复杂的,涉及多个基因表达的变化,还有大量未知靶点有待于进一步研究。 Objective:To investigate the effective target of Fuzheng Zengxiao Formula on lung adenocarcinoma ceils with genechip.Methods:Hairless mice were subcutaneous inoculated with PAa cells and divided into two groups.From the day of inoculation,the mice of group Ⅱ were given broth of Fuzheng Zengxiao Formula by intragastric administration,the mice of group Ⅰ with same volume of normal saline per day.When tumor diameter was about 1 centimeter,radiotherapy of β electron ray was given.Twenty three hours after radiotherapy,the mice was sacrificed and harvested the tumor quickly to get RNA from it.To analyze the difference of gene expression between the two groups with Affymetrix Human Genome U133 2.0 Array Gene Chips.Results:Eight hundred and fifty seven genes were differently expressed between groups,including 349 up-regulated genes and 508 down-regulated genes.Among them 39 genes were involved in GeneBank.Of them, 39 genes could be divided into cell cycle genes,apoptosis genes,signal transference genes and metabolism genes according to their function.Conclusion.Radiosensitization of Fuzheng Zengxiao Formula is complicated, many genes are involved,and a great quantity of unknown targets need to he investigated further.
