摘要 目的:建立以α_1-酸性糖蛋白柱(α_1-AGP)为手性同定相的高效液相色谱法直接拆分雷贝拉唑对映体。方法:研究了流动相中有机改性剂的种类和含量、流动相的 pH、缓冲溶液的浓度和流速等对拆分的影响。结果:以乙腈-0.02 mol·L^(-1)磷酸盐缓冲液(pH 6.0)(14:86)和甲醇-0.02 mol·L^(-1)磷酸盐缓冲液(pH 6.0)(20:80)为流动相,流速为0.8 mL·min^(-1),在5min 内实现了雷贝拉唑对映体在α_1-酸性糖蛋白柱上的拆分。结论:流动相中有机改性剂的种类和比例、流动相的 pH、缓冲溶液的浓度是影响拆分的重要因素。 Objective:To establish an HPLC method to separate rabeprazole and its enantiomer on α1-acid glycoprotein chiral column. Method: The effects of chromatographic conditions including pH of mobile phase, ionic strength,type and concentration of organic modifiers, flow rate on chiral separation were studied. Results: Rabe- prazole enantiomers were separated using an acetonitrile - 0. 02 mol ·L^-1phosphate buffer( pH 6. 0) ( 14: 86) and a methanol -0. 02 mol ·L^-1 phosphate buffer (pH 6.0) (20: 80) at 0. 8 mL·min^-1 Conclusion:The pH of mobile phase, ionic strength, type and concentration of organic modifiers,flow rate on chiral separation were important chromatographic variables in the separation of rabeprazole enantiomers.
