摘要
目的:建立河北道地药材知母的 HPLC-ELSD 指纹图谱分析方法,并与不同产地知母药材指纹特征相比较,为科学评价与有效控制知母药材质量提供新方法。方法:采用 HPLC-ELSD 法测定了道地知母等19个不同来源的知母样品。色谱条件:Diamonsil^(TM) C_(18)(250 mm×4.6 mm,5 μm)色谱柱,乙腈(A)-水(B)为流动相,梯度洗脱:0~20 min,23%A;25 min,31%A;35 min,33%A;55 min,48%A;60 min,48%A;65 min,23%A。ELSD 检测器,流速1.0 mL·min^(-1),柱温30℃。结果:建立了知母 HPLC-ELSD 指纹图谱共有模式,并对不同产地知母药材进行了相似度比较。结论:方法简便、可靠,为全面有效控制知母药材的内在质量提供了依据。
Objective :To establish a fingerprint analysis method for identification of Rhizoma Anemarrhenae of Hebei province,get the control chromatogram and compare the fingerprints of Rhizoma Anemarrhenae collected from different producing areas so as to establish a sensitive and specific method for controlling the quality of Rhizoma Anemarrhenae. Methods: The HPLC - ELSD fingerprints of Rhizoma Anemarrhenae from different places were obtained by Waters 1525 instruments. The HPLC separation was performed on a C18 analytical column(250 mm ×4. 6 mm,5 μm)gradient eluted with a mixture consisting of acetonitrile (A)and water(B) at a flow rate of 1.0 mL · min ^-1 with ELSD detector, using a linear gradient of 0 - 20 min ,23% A ;25 min ,31% A ;35 rain ,33% A ;55 min ,48% A ;60 min ,48% A ;65 min,23% A. The temperature of column was 30 ℃. Results: The mutual mode of HPLC - ELSD fingerprints was set up ,and the similarity degrees to the crude drugs of different producing areas were compared. Conclusions :The method is simple and reliable and can be used as a quality control item for Rhizoma Anemarrhenae.
