摘要
目的:在整个溶剂洗脱空间寻找最佳的分离途径,建立抗衰老中药复方的液相色谱指纹图谱测定方法。方法:100%水以10%为阶梯递减至0%,进行11次等度洗脱,在整个溶剂洗脱范围内,选取25个稳定响应,利用上述响应间的容量因子差之和作为分离优化指标函数,依据函数值最大走向的基本原则进行分离优化。结果:在有限洗脱时间内,实现了抗衰老复方指纹图谱分离条件的整体优化。结论:中药样品指纹图谱条件的优化方法不是一个最终优化模式的选择,而是在一定溶剂体系内稳定分离的一个局部最优条件的寻找,这需要在整个溶剂范围内寻找最佳分离路线,检测结果需要在整个检测范围内进行评价。结果同时表明对大量未知成分为主的复杂分析体系,通过简单的预试可以得到较好的分离,可为复杂分析样品的分离分析提供一个简便的方法。
Objective:The method intended to find an optimal elution path in whole solvent elution space for achieving the objective of integrated optimization with the assurance of local optimized process. Method:In order to obtain the variance of discernment responses' capacity factors within whole elution system and with the substance analysis of anti - aging compound recipe, it was established method of anti - aging compound recipe LC fingerprint chromatogram through 11 isocratic elution from 100% water phase to 0% water phase by using the sum of capacity factor differences as separation optimized function. Based on the principle of maximal function result to 25 stable responses, final optimized gradient elution way was selected as completed method and validated. Results:It achieved integrated optimizing separation of anti - aging recipe fingerprint within a limited running time. Conclusion: The optimization of LC chromatogram separation method is a key point for fingerprint acquisition. In the presentation of results ,TCM sample fingerprint chromatogram method optimization is not a selection of final optimized model, which is just a local optimized condition search in certain solvent system to obtain stable separation depended on studying in integral elution and detection scale. The results also presented that it could be obtained better separation with several simple pre -tests in the complex sample chromatogram analysis even with much unknown substance and provide a simple way to complex chemistry analysis.
