摘要
目的:建立超临界 CO_2萃取-反相高效液相色谱法测定三孢布拉霉菌超临界 CO_2萃取物中番茄红素含量的方法。方法:采用美国 Alltech Alltima C_(18)(4.6 mm×250 mm,5 μm)色谱柱,以甲醇-四氢呋喃-水(70:26:4)为流动相,流速为1.5mL·min^(-1),检测波长为472 nm。结果:线性范围为0.1~0.5 μg,相关系数 r=0.998;精密度试验的 RSD(n=7)为1.7%,加样回收率为98.3%~102.1%。结论:本文采用反相高效液相色谱法初步建立了三孢布拉霉菌超临界 CO_2萃取产物中番茄红素的定量方法,该法分析准确、快速,灵敏度高,重现性好。
Objective:To establish an RP - HPLC method for determination of lycopene in Blakeslea trispora extracts by supercritical carbon dioxide extraction. Methods:The content of lycopene in Blakeslea trispora extracts was determined by reverse phase HPLC at 472 nm on an Alltima C18 (4.6 mm× 250 mm,5 μm)column with methanol -THF -water(70: 26: 4)as mobile phase and flow rate 1.5 mL · min^-1. Results:The linear range for the content of lycopene was between 0. 1 and 0.5 μg,r =0. 998 ,the recovery was 98.3% to 102.1% and RSD of precision test was 1. 7 % ( n = 7 ). Conclusion: The method is simple, accurate and is suitable for the determination of lycopene.
