摘要 目的了解宁夏回族自治区流行麻疹野病毒的基因型。方法2004年5月,青铜峡市某小学发生麻疹爆发,收集了3份病人急性期咽拭子标本和11份血标本,分离咽拭子中的麻疹病毒,使用逆转录-聚合酶链反应检测分离到的麻疹病毒核蛋白(N)基因,对扩增后的N基因片段进行核苷酸序列测定,并分析鉴定基因型。结果宁夏回族自治区首次分离到2株麻疹病毒;酶联免疫吸附试验捕捉法检测病人血清的IgM抗体,阳性率为100%;序列分析结果表明,这2株麻疹病毒均属于H1基因型。结论2004年引起青铜峡市麻疹流行的病毒为H1基因型。 Objective In order to know the genotype of measles wild virus epidemic in Ningxia. Methods Three throat swab specimens and 11 blood specimens were collected from suspected measles cases of a school of Qingtongxia City, Ningxia in May, 2004. Using virus isolation method isolated MV from throat swab specimens. N gene of isolated MV strains was amplified by RT-PCR. The amplified N gene segment was sequenced and analyzed genotype. Results We isolated 2 MV epidemic strains in Ningxia at first time. Positive rate of MV-IgM antibody of patients detected by ELISA capture method was 100 %. Sequence analysis indicated that all the 2 epidemic strains belong to H1 genotype. Conclusion It was H1 genotype MV that caused measles outbreak in a school of Qingtongxia City, Ningxia in 2004.
