摘要
目的 观察不同浓度氢醌(HQ)对L-02肝细胞内POLH基因mRNA表达的影响,旨在探讨HQ对人体细胞毒性作用的分子机制。方法 应用四甲基偶氮唑盐比色法检测不同浓度(0、5、10、20、40、80、160和320μmol/L)HQ作用24h后对L-02肝细胞存活率的影响;应用带有SYBR GreenⅠ的实时荧光定量聚合酶链反应技术研究不同浓度(0、5、10、20、40、80和160μmol/L)HQ对L-02肝细胞内POLH基因mRNA表达的影响。结果 在0—80μmol/L的范围内。HQ对L-02肝细胞的存活率没有明显的影响(P〉0.05),当染毒剂量超过160μmol/L时,其存活率则明显地下降(P〈0.01);在HQ染毒剂量低于80μmol/L的范围内,POLH基因mRNA表达有随着剂量的增加而增加的趋势,当HQ的剂量达到160μmol/L时,POLH基因mRNA的表达则有所降低,但仍高于对照组。结论 HQ可以诱导L-02肝细胞内POLH基因在mRNA水平上的表达改变。
Objective To explore the effects of hydroquinone (HQ) on the expression of POLH mRNA in human L-02 hepatic cells, Methods MTT assay was conducted to detect the effects of HQ at various concentrations(0,5,10,20,40,80,160 and 320 μmol/L) on the survival rate of L-02 hepatic cells; real-time quantitative PCR with SYBR Green I technology was taken to measure the effects of HQ on the expression of POLH mRNA in L-02 hepatic cells exposed to HQ at various concentrations(0,5,10,20,40,80 and 160 μmol/L). Results MTT assay showed that HQ at the concentrations from 0 to 80 μmol/L had little effects on survival rate of L-02(P 〉0. 05) ; whereas the survival rates in both groups of 160 and 320 μmol/L were significantly higher than that in the control (P 〈0.01 ) after treated with HQ for 24 hours. It was found that HQ at low concentrations ( 〈 80 μmol/L) could induce increased expression of POLH mRNA which was consistent with the increase of HQ concen- trations; the expression of POLH mRNA decreased once the concentrations of HQ exceeded 160 μmol/L when compared with that of the group 80 μmol/L,but it was higher than that of the control. Conclusion This study suggests that HQ might induce changes of the expression of POLH mRNA in hepatic cells.
