摘要 目的:筛选和优化A群脑膜炎多糖的衍化工艺。方法:将A群奈瑟氏脑膜炎球菌多糖经溴化氰活化后通过己二酰肼衍生后制备出适宜的多糖衍生物。对衍化条件进行摸索和优化后通过衍化率及衍生物的可溶性来考察衍化效果,同时通过生化指标来考察该衍生工艺制备的结合疫苗的质量和收率。结果:在pH 11.0±0.2、活化30 min以及溴化氰与多糖比值(w/w)=0.5时,衍生物制备的结合疫苗各项生化指标在所筛选的条件中最优。结论:经过筛选优化的衍生工艺提高了多糖的衍化率,适宜结合疫苗的研制,保证了其结合疫苗的免疫原性。 Objective: To compare and optimize the craft of derivation of serogroup A meningococcus polysaccharicde. Methods: The serogroup A Neisseria meningococcus polysaccharide which had been activated by cyanogen brom/de(CNBr) was combined with 1,6 - adipic acid dihydrazide(ADH) and the eligible polysaccharide derivation was obtained. After the and optimization for the several key conditions of the processing, the derivation rate, solubility of the derived polysaccharide and some biochemical properties of the conjugate vaccine were examined in order to determine the quality and yield of the conjugate vaccine. ResultS:Under the condition of ph 11.0i0. 2, activation for 30 rain and cyanogen bromide/polysaccharide= 0. 5, the biochemical criterions of the nmnu[actured conjugate vaccine was excellent. Conclusion:The polysaccharide derivation rate is not only promoted by the chosen parameters of derived processing, but also the immunogenicigy of conjugate vaccine is well ensured.
