摘要
目的研究银杏内酯对冈田酸(OA)诱导拟AD大鼠行为学和海马CA1区bcl-2表达的影响。方法大鼠海马CA1区多次微量注射OA,建立拟AD动物模型。部分模型动物同时用银杏内酯灌胃。Morris水迷宫实验检测大鼠学习记忆能力,免疫组化方法观察大鼠海马CA1区bcl-2表达。结果与拟AD模型组相比,银杏内酯组水迷宫定向航行试验平均潜伏期明显缩短(P<0.05或P<0.01);空间探索试验原站点象限活动时间明显延长(P<0.05),站台穿越次数明显增多(P<0.05)。大鼠海马CA1区bcl-2阳性表达明显增多(P<0.05)。结论促进海马CA1区bcl-2表达上调,从而减少神经细胞的凋亡是银杏内酯改善拟AD大鼠学习记忆的可能机制之一。
Objective To investigate the effect of the gingkgolide treatment on the ethology and the expression of bcl-2 in the hippocampal CA1 area of the Alzheimer' disease (AD) like model rats. Methods Oksdaic acid (OA) was injected into hippocampal CAI area of rats with microdosis repeatedly to establish Alzheimer' disease-like model. Gingkgolide was intragastric adminstraetion in the treatment rats at: the same time. The learning and memory abilities of rats were examined by means of Morris Water Maze, and immunohistochemistry were employed to observe the expression of bcl-2. Results Compared with ADlike model rat, gingkgolide group had short average escape latency in place naviga- tion test(P〈0. 05, P〈0. 01) ; and longer swimming time in the third quadrant in spatial probe test after the platform was removed (P〈0. 05). The expression of bcl-2 in the hippoeampal CA1 area of the AD-like model rats were significantly increased. Conclusion Gingkgolide can affect the up-regulation of the expression of bcl-2 in the hippocampal CA1 area, then inhibit the apoptosis of neurons. This may explain one of mechanisms by which gingkgolides enhance learning and memory.
