摘要
对来自大兴安岭、长白山的高山红景天花粉进行了超低温保存研究。结果表明:不同的干燥时间对超低温保存后花粉萌发力有明显影响,12 h干燥处理的花粉萌发力明显高于8 h、24 h干燥处理的;用0℃(8 h)→-10℃(8 h)→-20℃(8 h)变温预冻处理能大幅提升超低温保存后花粉的萌发力;超低温保存后的解冻方式以-20℃(8 h)→4℃(8 h)→25℃(8 h)为最佳。
Researches had been carried out on cryopreservation of pollen from Changbai Mountain and Daxinanling Mountain. The results indicated that desiccation duration affected the pollen germination after cryopreservation. The germination Viability of pollen that was prefrozen for 12 h was higher than that of 8 h, 24 h. Treatment at varied temperate of 0℃ (8 h)→-10℃(8 h)→-20℃(8 h)could greatly increase the viability of pollen stored in liquid nitrogen before being thrown into liquid nitrogen. The defreezing method of-20℃ (8 h) →4℃ (8 h→25℃ (8 h)was better than other two treatments.
