槐果碱对人红白血病K562细胞的增殖抑制和诱导凋亡作用 被引量:13

摘要 研究了槐果碱对人红白血病细胞株K562的增殖抑制和诱导细胞凋亡的作用,应用流式细胞仪进一步观察槐果碱对K562细胞周期的影响。结果表明:槐果碱对K562细胞生长有明显的抑制作用,且这种抑制作用呈浓度依赖性、时间依赖性。细胞经0.8 g/L、1.0 g/L的槐果碱作用48 h后,即呈现典型的细胞凋亡形态学变化,作用72h后DNA凝胶电泳出现片段的梯形条带。流式细胞仪分析显示,用1.0 g/L槐果碱使K562细胞阻滞于G0/G1期,作用72 h后G0/G1细胞比例由38.0%上升到56.2%。槐果碱对人红白血病细胞K562的增殖具有明显的抑制作用,并能诱导其发生凋亡。 Effects of sophocarpine on the proliferation and apoptosis of human eryhroleukemia K562 cells were studied in this paper. Cell cycle distribution induced by sophoearpine was further analyzed by flow cytometry (FCM). Sophocarpine could inhibite the proliferation of K562 cells with concentration dependent and time dependent manners. When incubated with sophocarpine (0.8 g/L,1.0 g/L) for 48h, K562 ceils showed morphological changes associated with the characters of apoptosis under fluorescent microscope. Typical DNA ladder was found in agarose gel electrophoresis when K562 ceils were exposured with sophocarpine (0.8 g/L,1.0 g/L) for 72 h. The outcome of FCM showed that K562 ceils were arrested significantly in Go/G1 phase when treated with 1,0 g/L sophocarpine for 72 h, which increased from 38.0% to 56.2%. Sophocarpine can inhibit the proliferation of K562 and induce the apoptosis of the ceils.
