人血清白蛋白基因的序列特征分析

摘要 目的克隆人血清白蛋白(HSA)基因并对其序列进行分析。方法利用PCR方法从人胎肝cDNA中克隆HSA基因片段,用DNASIS(2.5)软件分析核苷酸及其推导的氨基酸序列,用SignalP3.0Server预测N末端氨基酸信号肽,用ClustalX(1.81)和GeneDoc对蛋白序列进行多重序列联配分析。结果成功克隆HSA基因,有15个外显子和14个内含子,与猴、猫、牛、兔和老鼠的蛋白氨基酸相似率分别为93%、82%、76%、75%、73%;含有3个保守的白蛋白结构域。结论本研究为开发长效蛋白药物奠定了良好的基础。 [Objective] To clone the human serum albumin(HSA) gene and analyze its sequence. [Methods] A DNA fragment of 1 884 bp was cloned by PCR from human fetal liver cDNA. Sequencing was analyzed by DNASIS(2.5) software. N-terminal signal peptide was predicted by SignalP3. 0 Server. And the protein multiplesequence was analyzed by Clustal X (1.81) and GeneDoc. [Results] The HAS gene was cloned successfully: Analysis of genomic structure by comparing it with human genomie sequence revealed that it contained 15 exons and 14 introns. The deduced amino acid sequence of HSA showed similarities of 93%, 82%, 76%, 75% and 73% with those from Macaca mulatta, Fells catus+ Bos taurus, Oryctolagus cuniculus and Rattus norvegicus. Bioinformatical analysis indicated that HSA was composed of three conserved albumin domains. [Conclusion] This study provides a base for improving long half-life of protein drug.
