摘要 目的建立兔眼视网膜母细胞瘤(Rb)动物模型,便于和扩展应用眼肿瘤动物模型进行临床实验治疗。设计实验性研究。研究对象8只兔(16眼)及HXO-Rb44细胞株。方法全麻下在兔眼脉络膜上腔注入浓度为(1.17×107)/ml肿瘤细胞悬液0.1ml,分别于种植后的l、3、7、14天观察记录兔眼底改变,第14天摘除眼球后行病理观察和肿瘤细胞鉴定。主要指标观察肿物生长数量及生长方式,并以病理切片H-E和免疫组化染色证实细胞来源。结果种植后1周肿物开始生长,生长方式有肿块状和弥漫状,成活率为7/16。肿瘤细胞生长活跃,生长旺盛区与坏死区相间隔;细胞呈未分化的特点:核大深染,胞浆少,核与胞浆比例失调,异形性明显,并见大量的核丝分裂。经H-E和免疫组化染色,证明兔眼Rb的生长方式、分化类型及细胞特征与人眼Rb相一致。结论HXO-Rb44细胞株可以在兔眼内成活,并能形成实体性肿瘤,兔可以成为较理想的Rb模型动物。 Objective To establish the animal model of retinoblastoma in the rabbit eye, in order to extend the experimental clinical ophthalmic therapy on the tumor of animal model. Design Experimental study. Participants 16 eyes from 8 rabbits, the tumor cell line is HXO-Rb44. Method 0.1ml of the cell suspension with the concentration of (1.17×10^7)/ml were injected into the choroid of rabbits after general anesthesia; exam and record the ocular fundus of the rabbits after 1, 3, 7, 14 days of implantations, and enucleate the rabbits eyeballs for pathologic examination and tumor cell identification at 14th day. Main Outcome Measures The quantity and the type of neoplasm, verified by pathologic examination with H-E stain and immune chemical stain. Results Neoplasm was seen growing in the rabbit eyes one week later after implantation, in shape of lump or widespread; the survival rate is 7/16. The tumor cells grow actively with evident interval regions of growing and necrosis. The tumor cells have undifferentiating features: bigger and deeply stained nucleus, less cell plasma, disorder nucleus and plasma ratio, dense chromatin, morphology very different and many mitosis. In rabbit Rb model it is testified that the growth mode, differentiation type and cell characters are as same as those in human Rb with H-E and immune chemical stain. Conclusion Cell line HXO-Rb44 can be alive and form tumor in the rabbit eyes. Rabbit may be an ideal ani mal for Rb model.
