摘要
目的探讨通心络、辛伐他丁对兔颈动脉粥样硬化性斑块ABCA1、RXRα表达的影响,并评价各自的优缺点。方法将32只新西兰大白兔分为4组,A组:正常对照组8只,余24只建立颈动脉粥样硬化性狭窄大白兔模型,并分为B组:模型对照组8只;C组:辛伐他丁组8只(辛伐他丁5mg.kg-1.d-1,1次.d-1);D组:通心络组8只(通心络1g.kg-1.d-1,1次.d-1);药物干预前后检测兔静脉血的TG、TC、LDL及HDL水平,药物干预2周后用WesternBlot法检测右侧颈动脉狭窄段血管ABCA1、RXRα蛋白表达量。结果通心络、辛伐他丁干预组较模型组、正常对照组ABCA1、RXRα蛋白表达均上调(P<0.05),血脂水平较模型对照组下降(P<0.05);辛伐他丁组较通心络组ABCA1、RXRα蛋白表达上调、血脂下降程度更明显(P<0.05);但辛伐他丁组较通心络组ALT增高(P<0.05)。结论通心络、辛伐他丁可使兔颈动脉粥样硬化斑块ABCA1、RXRα蛋白表达水平上调,这可能是两种药物抗动脉粥样硬化斑块形成的机理之一;辛伐他丁在降脂、上调ABCA1、RXRα蛋白表达方面好于通心络,但通心络无损害肝功能的副作用、而辛伐他丁损害肝功能的副作用明显。
Objective To investigate the effects of TONGXINLUO capsule and simvastin on the expressions of ATP-binding cassette transporter A1 (ABCA1) in the rabbit carotid artery atherosclerotic plaque. Methods 32 Male New Zealand white rabbits were randomly divided into four groups. Each group included 8 rabbits. A group(blank). The positioning of a improved shape of silicone collar around the carotid artery of the 24 rabbits that were fed with hypsocholesterol diet for 2 weeks to build the experimental carotid artery stenosis models. B group: carotid artery stenosis control. C group and D group: the models were subsequently treated with simvastin (5 mg·kg^-1 ·d^- 1 ) TONGXINLUO capsule 1g·d^-1 respectively for 2 weeks. The rabbit carotid artery atherosclerotic plaque was excised after treatment for 2 weeks, the protein levels of ABCA1 and RXRα were detected by Western blot, respectively. Detected the contents of TG, TC, LDL-C, HDL-C and ALT in blood before and after treatment with drugs. Results After treatments with TONGXINLUO capsule and simvastin,the protein expression levels of ABCA1 and RXRα all increased,and the contents of blood-fat decreased (P〈0.05),C group was more significant than D group (P〈0. 05), but the level of ALT was significantly higher in C group than in D group(P〈0.05). Conclusions TONGXINLUO capsule and simvastin can up-regulate the protein expression levels of ABC1 and RXRα. These could be the one mechanism of them resisting the carotid artery atherosclerotic plaque. In these parts simvastin is better than TONGXlNLUO capsule,but it's side effect to liver is more significant than TONGXlNLUO capsule.
