基质金属蛋白酶-12及其编码基因NM_002426在喉鳞状细胞癌中表达的研究 被引量:7

摘要 目的:研究基质金属蛋白酶-12(MMP-12)蛋白及其基因片段在喉鳞状细胞癌中的表达情况及其与喉癌生物学行为的关系。方法:选取30例声门上喉癌新鲜组织标本(实验组)及0.5cm外正常黏膜组织(对照组),按标本恶性度低、中、高度不同,分别制作不同恶性度的基因芯片,研究其上游基因NM-002426表达比率;用免疫组织化学的方法检测其在组织中的表达情况。结果:NM-002426基因在3种不同恶性度基因芯片中都为高表达比率,较同族的基因片段比率均高,在中、高度恶性芯片中的比率是相应芯片中测定结果中的最高值;免疫组织化学结果显示实验组与对照组MMP-12表达率差异有统计学意义(P<0.01),实验组中转移者与未转移者表达差异有统计学意义(P<0.05);而不同恶性度、不同性别间差异无统计学意义。结论:在喉鳞状细胞癌的发生、发展中,NM-002426基因显著增殖,编码产生的大量MMP-12对肿瘤的转移起到了关键作用;MMP-12及其基因的高表达可作为喉癌的辅助诊断依据。 Objective: Matrix metalloproteinases (MMPs) are key factors which influence the grades and/or metastasis of stereo turnout. There are short of research of MMP-12 now. The experiment is done to investigate the expression of MMP-12 and NM_ 002426, and relationship between the two genes and the biologic behavior in squamous laryngeal carcinoma. Method: Thirty pairs of specimens, include supraglottie carcinoma tissue and normal mucosa, were choo.sen, Three gene arrays, based on different kinds of malignant, were made to study NM_ 002426 at first, then immunohistochemical method were used to observe the difference of MMP-12 between tumour and normal mucosa, metastasis group and no metastasis group. Result: NM_ 002426 express high ratio in all 3 chips, and have the highest mark in both mid level and highqevel malignanl tissue. The immunohistochemical results showed that MMP 12 had significant difference between tumour and normal mt, cosa, metastasis group and no metastasis group. The MMP-12 expression was unrelated to different malignant degree. Conclusion: NM_ 002426 has significant byperplasy in squamot, s laryngeal carcinoma, which caused high expression of MMP 12 ,and it makes a main contribution to the carcinogenesis and metastasis of laryngeal carcinoma. MMP 12 and its gene overexpress may be a auxiliary diagnosis method.
