共固定化肿瘤坏死因子和干扰素的协同抗癌作用 被引量:5

摘要 研究重组人干扰素(IFN-γ)和重组人肿瘤坏死因子(TNF-α)的共固定化对体外培养的人宫颈癌细胞(Hela)生长的影响。利用光固定化方法将IFN-γ、TNF-α共固定在24孔聚苯乙烯培养板(PSt)上,培养Hela细胞,分别设为共固定(IFN-γ+TNF-α)、单固定(IFN-γ、TNF-α)、游离(IFN-γ、TNF-α、IFN-γ+TNF-α)各组培养,研究癌细胞生长抑制曲线,透射电镜、流式细胞仪检测Hela细胞的增殖和凋亡。结果表明共固定化干扰素和人肿瘤坏死因子对Hela细胞的抑制作用显著,最高达82%,显示IFN-γ对TNF-α具有良好的协同抗宫颈癌作用。 To explore the effect of the co-photo-immobilization of Recombinant Human Interferon-r (IFN-γ) and Recombinant Tumor Necrosis Factor (TNF-α) on the Hela cell cultivated in vitro. Co-immobilize the photoactive IFN-γ and TNF-α on the polystyrene membrane ; Cultivate the Hela cell on the cell culture polystyrene plate surface and set up the cultivation of the photo-immobilization IFN-γ+TNF-α,Photo-immobilization IFN-γ,nature (IFN-γ,TNF-α,IFN-γ+TNF-α). After 48 hours, the inhibition effect against Hela cells was studied in vitro by SEM and flow cytometry. It showed that the co-immobilization IFN-γ+TNF-α has a more obvious inhibition effect on the Hela cell. And IFN-T can cooperate with TNF-α obviously.
