摘要
利用重组工程细胞进行基因治疗无疑为多种难治性疾病的有效治疗提供了新的思路,但宿主对异源细胞产生的移植排斥反应却是其在临床上应用的主要障碍。现代细胞微囊技术因其具有良好的免疫隔离屏障作用,使克服这一障碍成为可能。近年来,细胞微囊技术在基因治疗方面的发展迅速,已在血友病,帕金森病、肿瘤等多种难治性疾病的治疗应用中显示出广阔的应用前景。本文就细胞微囊技术的微囊膜材料、制作工艺、微囊内重组细胞的构建以及其在基因治疗方面的研究作一综述。
It is no doubt that the gene therapy using recombinant engineering cells provides a novel approach to many refractory diseases. However, the transplant rejection from the host's immune system against heterogeneous cells has been the main handicap of its clinical application. The modern cell micro-encapsulation technique with good immune isolation makes it possible to overcome this problem and has shown potential application foreground in clinical therapies for a lot of diseases such as Parkinson's disease and Hemophiliac disease. This article reviews mainly the relative materials and techniques in processing mlcro-encapsulation, the host cells used to construct the recombinant genetic engineering cells and application of cell micro-encapsulation technique in the field of gene therapy.
