醉酒灵Ⅱ号药效学研究及作用机理探讨 被引量:2

摘要 目的探讨中药复方“醉酒灵Ⅱ号”对急性酒精中毒小鼠血中乙醇浓度、肝中乙醇脱氢酶(al-cohol dehydrogenase,ADH)活性和脑组织中单胺神经递质的影响。方法雄性小鼠随机分为模型组和醉酒灵Ⅱ号高、中、低剂量组,模型组与醉酒灵Ⅱ号高、中、低剂量组灌酒0.01 mL/g,复制急性酒精中毒动物模型,0.5 h后给药组给予不同浓度的醉酒灵Ⅱ号,给药后用色谱法测定脑组织中单胺神经递质含量;用分光光度法测定血中乙醇浓度和肝中ADH活性。结果醉酒灵Ⅱ号组酒后1.5 h和2.5 h的血中乙醇浓度明显降低,与模型组比较有显著性差异(P<0.05)。醉酒灵Ⅱ号组酒后2.5 h肝中ADH活性虽与模型组比较无显著差异(P>0.05),但有增强趋势。醉酒灵Ⅱ号组酒后1 h脑组织中去甲肾上腺素(NE)、3,4-二羟基苯乙酸(DOPAC)含量明显增加,与模型组比较有显著性差异(P<0.05,P<0.01);HVA含量明显减少,与模型组比较有显著性差异(P<0.05)。结论醉酒灵Ⅱ号可降低急性酒精中毒小鼠血中乙醇浓度,改善急性酒精中毒时小鼠脑组织内单胺递质的变化,具有一定解酒和对抗酒精对神经系统的毒性作用。 Objective activity of alcohol deh of mouse model with To observe the effect of Zuijiuling Ⅱ on the alcohol concentration in blood, the ydrogenase (ADH)in the liver and the concentration of neurotransmitter in the brain acute alcoholism. Methods Female mice were randomly divided into 4 groups including model group, high-dose Zuijiuling Ⅱ group, middle-dose Zuijiuling Ⅱ group and low-dose Zuijiuling Ⅱ group. Mouse model with acute alcoholism was duplicated by feeding the mice with Site Wine at the dose of 0. 01 mL/g. In Zuijiuling Ⅱ groups, Zuijiuling Ⅱ was given by intragastric administration 0.5 hours after alcohol feeding. Stratography was used to detect the content of some neurotransmitters in brain and spectrophotometric method was adopted to determine the alcohol concentration in blood and the activity of ADH in the liver. Results Compared with that in the model group, the alcohol concentration in blood in Zuijiuling Ⅱ groups was decreased significantly 1.5 hours and 2.5 hours after alcohol feeding (P 〈 0.05 ) and the activity of ADH in the liver had tendency of increase 2.5 hours after alcohol feeding (P 〉 0.05 ). The content of norepinephrine (NE) and dihydroxy - phenyl acetic acid (DOPAC) in the brain 1 hour after alcohol feeding was increased (P 〈0. 05, P 〈0.01 ), while the content of HVA was decreased(P 〈 0. 05) compared with that in the model group. Conclusion Zuijiuling Ⅱ can reduce the alcohol concentration in blood of mouse model with acute alcoholism and increase the content of NE and DOPAC. It has effect on relieving alcohol toxicity and antagonizing toxic effect of alcohol on nervous system.
