柴胡-黄芩水煎液中不同化学成分群配伍与其解热作用相关性研究 被引量:14

Correlation between compatibilities of different chemical component groups in Chaihu-huangqin Decoction and their antipyretic effects

摘要 目的 探讨柴胡-黄芩水煎液中分离出的5类主要化学成分群的不同配伍与解热作用的相关性,旨在从化学成分群层次上揭示柴胡-黄芩药对解热的药效物质基础。方法 依照正交设计L8(2^7)表将柴胡-黄芩合煎液中分离出的5个化学成分群,依次配制成化学成分群配方1-8。以80μg/kg剂量一次性腹腔注射LPS复制大鼠发热模型;中药各组分别灌胃给予化学成分群配方1-8(剂量均为折合原药材10g/kg);从注射LPS开始,分别于30、90、150、210、270、330min测量体温,连续观察330min,计算各组体温变化值△T。结果 在大鼠发热第2、3峰值处(150、330min),配方1、5、6组能够显著降低LPS引起的大鼠体温升高(P〈0.05),显示出明显的解热作用。与配方1组相比,各组间不存在显著性差异,但配方5、6组具有更强的解热趋势。各因素方差分析结果表明,化学成分群挥发油部分(B)、黄酮部分(D)和剩余混合物部分(E)在解热效应中发挥重要作用,能协同增强解热效果;而皂苷部分(C)具有明显的拮抗作用;多糖部分(A)对解热作用无显著影响。结论 柴胡-黄芩药对中化学成分群的不同配方对发热模型表现出不同的作用,表明各化学成分群的不同配伍与解热作用的相关程度不同。化学成分群BDE为正交设计法优选出的抑制LPS诱导大鼠发热的最佳配方。 Objective To discuss the correlation between the compatibilities of 5 chemical component groups separated from Chaihu-huangqin Decoction and their antipyretic effects in order to reveal the substance basis of antipyretic effect of Radix Bupleuri and Radix Scutellariae at the level of chemical component group. Methods According to the orthogonal design table L8 (2^7), eight chemical component formulae were compounded from 5 chemical component groups separated from Chaihuhuangqin Decoction. The fever model was established by injecting LPS once intraperitoneally in the dose of 80 μg/kg in rats. The above-mentioned 1 to 8 formulae were given orally to 8 rat groups respectively in the dose of 10 g/kg (according to the herbal material). The rat rectal temperature was measured 30, 90, 150,210,270 and 330 minutes after the injection of LPS. The change values of the rectal temperature (AT) in different rat groups were calculated. Results The higher rectal temperature of rat induced by LPS in group 1, 5 and 6 were significantly decreased ( P 〈 0. 05 ) at 150^th and 330^th minute with a significant antipyretic effects. There was no significant difference between other groups and group 1, buta stronger trend of antipyretic effect in group 5 and 6. Analytic result of variance of all factors showed that group B (volatile oil), D (flavonoid) and E (other component mixture) in chemical component groups played important role in antipyretic effect with synergetic effect, and group C (saponin) showed a significant antagonisis, and A (polysaccharide) showed no significant effect. Conclusion The different compatibilities of chemical component groups in Chaihu-huangqin Decoction have different effects on rat fever model, which suggests that there are different correlations between different chemical component groups and antipyretic effects. The compatibility of group B, D and E is the best formula selected by the orthogonal design method for fever induced by LPS in rAt~
