摘要
目的通过口服丹参饮片大鼠体内化学成分指纹图谱的研究,探索丹参饮片口服产生疗效时有效物质基础成分的动态变化过程。方法按照中药有效物质基础研究方法学路线,采用HPLC色谱指纹图谱技术和体内药物分析方法。结果丹参饮片水煎口服后,发现16种以色谱峰代表的化学成分通过消化道吸收入血,其中4种为原型入血成分,12种为肠代谢后转化的新成分;血液中运载成分与入血成分相比,入血成分经过肝代谢后中有4种含量降低;经过肾代谢的尿液中有与血液运载相同的成分;心脏和脑组织中未发现成分进入。结论按照中药有效物质基础研究方法学路线,丹参饮片口服用药产生疗效的有效物质基础化学成分动态变化过程得到了揭示。
Objective To explore the dynamic course of basic chemical constituents of effective substance through the analysis of fingerprint chromatograms of chemical compositions in body fluid of rats after taking Radix Salviae Miltiorrhizae decoction orally. Methods According to the methodology of study on effective constituents in traditional Chinese drugs, HPLC fingerprint chromatograms technique and analysis method of drug in vivo were adopted. Results Sixteen kinds of chemical constituents, which were presented by chromatogram peaks, were absorbed into blood via alimentary tract after oral administration of Radix Salviae Miltiorrhizae. Among them 4 kinds were prototype and 12 kinds were new constituents transformed by intestinal metabolism. Four kinds of chemical constituents decreased after liver metabolism. Some chemical constituents in urine after kidney metabolism were same as those in blood. There was not any chemical constituents were found in brain and heart. Conclusion According to the methodology of study on effective constituents in traditional Chinese drugs, the dynamitic course of basic chemical constituents of effective substance of Radix Salviae Miltiorrhizae after oral administration has been revealed .
