摘要
目的研究细胞凋亡在子宫肌瘤组织射频治疗中的作用。方法光镜和电镜检测多电极射频消融的子宫肌瘤组织及未消融的子宫肌瘤组织中的凋亡细胞。用免疫组化二步法检测20例多电极射频消融的子宫肌瘤组织及未消融的子宫肌瘤组织中Bc l-2(B cell lymhpom a/leukem ia-2)和Bax(Bc l-2 assoc iated x-prote in)蛋白的表达。结果在消融灶边缘及其外0.5 cm的子宫肌瘤组织,细胞凋亡指数(16.3±3.4)高于未消融的子宫肌瘤组织(7.1±1.6,P<0.01);同时Bc l-2蛋白表达减弱(P<0.05),Bax蛋白表达增强(P<0.05)。结论高温诱导的细胞凋亡在射频消融致子宫肌瘤治疗中起着相关作用,可能是消融引起肌瘤坏死的机制之一。
Objective To detect the expression of apoptosis gene Bcl-2 and Bax in ablated hysteromyoma tissues, and to approach the principle of thermal therapy by radio-frequency. Methods Apoptosis cells in ablated hysteromyoma tissues and unablated were detected with microscope and electron microscope. Specimens from the margin of necrosis focus and its locus 0.5cm far from the margin were stained by HE. In randomly chooscn 10 high power fields, average apoptosis cell and apoptosis index were counted. The expression of Bcl - 2 and Bax protein of 20 hysteromyoma patients were determined in ablated and unablated tissues with two- steps immunohistochemical test. Results In ablated tissues and the site of O. 5 cm far from the margin, apoptosis index in ablated tissues was (16.3 ± 3.4 ) . higher than that in unablated tissues (7.1 ± 1.6) (P 〈 0.01 ). Compared with unablated myomas, the expression of Bcl-2 protein in ablated myomas was weak ( P 〈 0.05 ) and the expression of Bax protein in ablated myomas was strong ( P 〈 0.05 ). Conclusions Cell apoptosis is induced by high temperature relating with the myoma destroy after radio-frequency ablation. Perhaps apoptosis is one of the mechanisms of myoma necrosis induced by radiofrequency ablation.
