摘要 采用RAPD方法,对24个来自贵州代表性小麦白粉菌Blumeria graminis f.sp,tritici的单孢培养物进行生物学特性指纹分析,并与传统生理小种鉴定结果相比较,结果表明,在筛选的30个引物中,每个引物对代表菌株分别扩增出3—7条带,选用条带比较明显的5条引物,扩增得到的39个RAPD位点,主要集中在分子长300—2500bp,其中28个有多态性,多态性位点频率占71.8%。表明贵州小麦白粉菌遗传多态性丰富。从遗传距离的角度看.24个小麦白粉菌菌株的遗传距离变化幅度较大,分布在0.1719—0.8044。经0~1聚类分析,相同生理小种并不聚为一类,即小麦白粉菌毒性多态性与DNA多态性不形成对应关系,同一地域内的菌株在聚类图中近缘性更为紧密。 24 strains of Blumeria graminis f. sp. tritici, which were separated from representative areas or belonged to different races, were analyzed with RAPD ( random amplified polymorphism DNA ) method, The results were compared with the results of the usual method of identifying physiological races. During the test, 30 primers were selected and everyone showed significantly different lines from 3 to 7.39 loci were obtained through amplication with 5 primers, 71.8% with 28 polymorphic loci were polymorphic frequence. Loci focused on 300 -2 500bp,24 isolates were tested with the genetic distance from 0.171 9 -0.804 4. According to 0 - 1 statistic analysis, clustering analysis showed that the same physiologial race collected untogether, and the relation of strains coming from the same area was closer in linetrees.
