活血益气方对大鼠颈动脉球囊损伤后增殖细胞核抗原及细胞周期蛋白依赖性激酶表达的影响

摘要 目的为探讨活血益气方干预球囊损伤大鼠颈总动脉内膜增殖的机制,对损伤血管细胞核增殖抗原(PCNA)及细胞周期蛋白依赖性激酶(CDK2,CDK4)表达进行观察。方法随机将18只雄性SD大鼠分为假手术组、模型组、活血益气方组3组,球囊损伤大鼠颈总动脉前1d始给假手术组与模型组0.8mL/(100g·d)生理盐水灌胃,用药组给予相同剂量的中药灌胃,连续至第5天处死大鼠取其损伤的颈总动脉,用免疫组织化学方法检测PCNA、CDK2、CDK4蛋白的表达。结果各组PCNA、CDK2、CDK4均表达阳性,假手术组有极少量表达,模型组明显增多,用药后表达量显著减少,与模型组比较有统计学意义(P<0.01)。结论活血益气方对球囊损伤后动脉血管内膜增生的抑制作用机制可能与下调细胞核增殖抗原及细胞周期蛋白依赖激酶的表达,抑制血管内膜平滑肌细胞的增殖有关。 Objective To investigate the effect of Huoxueyiqi drug on expression of proliferation cell nuclear antigen(PCNA) and cyclin- dependent kinase (CDK2, CDK4) after carotid artery vascular injury in rats. Methods Rats( n = 18) were randomly divided into 3 groups: Group 1 ( sham - operation), group 2 (vascular balloon injury) and group 3 [0.8 mL/100(g/d) of Huoxueyiqi was given after injury]. The expression of PCNA, CDK2 and CDK4 was observed by immunohistochemical technique. Results The expression of PCNA, CDK2 and CDK4 at the 5th day after injury was maximum in group 2 (model group), and reduced obviously when given Huoxueyiqi, compared with group 1 and group 2 ( P 〈 0.01 ). Conclusion Huoxueyiqi could effectively decrease intimal proliferation after balloon injury, which might be associated with smooth muscle cell through reducing CDK4. inhibiting the proliferation of vascular the expression of PCNA, CDK2 and CDK4.
