运用生物信息学技术分析预测抗氨基末端脂多糖结合蛋白(NH-LBP)单链抗体的结构

摘要 应用生物信息学技术预测人工构建的人源化抗氨基末端脂多糖结合蛋白(NH-LBP)单链抗体(scFv)的结构及其生物学活性。利用在线生物信息学工具分析抗NH-LBP的scFv的理化特性及该抗体的二、三级结构。预测结果表明抗NH-LBP的scFv属于较稳定蛋白质,体内半衰期较长;该抗体与数据库中收录的单链抗体的二级结构十分相似;该抗体的三维空间结构为一种球形的蛋白质。通过在线生物信息学工具获得了抗NH-LBP scFv的一、二、三级结构及理论功能,为进一步研究该抗体的体内外生物学活性提供了理论依据。 To predicate the structure function relationship of single chain fragament variable of antibody against NH2-terminal lipopolysaccharide binding protein, the physico-chemical properties of scFv against NH-LBP were analyzed by bioinformatics methods, and its secondary structure and tertiary structure were predicted too. The scFv against NH-LBP is a stable protein and has long half-life in vivo, and its structure is similar to the published structure of single chain variable fragment antibody and its 3-D structure is globular just like that of antibody. The physico-chemical properties, structure information and theoretical bio activity of scFv against NH LBP were obtained By informatics methods online. It provides the theoretic foundation to study the bio-activity of this antibody.
