摘要
用纯化的人结缔组织生长因子(connective tissue growth factor,CTGF)免疫BalB/C小鼠,采用杂交瘤技术制备CTGF单克隆抗体(monoclonal antibody,McAb),获得2株稳定分泌抗体的杂交瘤细胞系7C7和8A11。两株细胞染色体众数分别为95条和91条;单抗亚类鉴定结果显示2株单抗均为IgG1型;间接ELISA法测定7C7和8A11腹水效价分别为:1.3×105和8.1×104;相对亲和力:7C7>8A11。CTGF单克隆抗体的制备为相关实验研究和临床诊断提供了条件。
BalB/C mice were immunized with purified CTGF, and McAb was prepared by hybridoma technique. Two hybridoma cell lines (7C7,8A11) secreting the McAb against CTGF were obtained. The quantity of chromosome in 7C7 and 8A11 is 95 and 91 respectively. Ig subgroups of 7C7 and 8A11 are IgG1. The titers of ascites in 7C7 and 8A11 were 1.3 × 10^5 and 8. 1 × 10^4 respectively. Relative affinity of 7C7 is higher than that of 8A11. Preparation of McAb against CTGF provides the precondition for relative research and clinical diagnosis.
