三硝基苯磺酸诱导大鼠溃疡性结肠炎的实验研究 被引量:26

摘要 目的探讨三硝基苯磺酸溃疡性结肠炎模型大鼠NO、MDA、SOD的动态变化。方法38只SD大鼠留取6只作正常对照组,剩余32只大鼠按造模后处死取材顺序依次分为5个组:第1天(6只)、第3天(7只)、第7天(6只)、第14天(6只)、第21天(7只),观察大鼠结肠长度、溃疡分数,检测结肠上清NO、MDA、SOD。结果各造模组大鼠结肠缩短以第7天组最为明显,与正常组比较(P<0.01),其余各组(P<0.05);溃疡分数以第7天组增加最为明显,与正常组比较(P<0.01),其余各组(P<0.05)。结肠上清第1天、第3天组的NO较正常组明显增加(P< 0.01)、(P<0.05);第1、3、7天的MDA呈渐增趋势,其中第3天、第7天组与正常组比较(P<0.05);SOD活力较正常组不同程度降低,但差异均无显著性意义(P>0.05)。结论TNBS所诱导溃疡性结肠炎模型大鼠局部溃疡分数,结肠上清NO、MDA呈现先升后降的成模及自愈趋势,符合溃结研究模型所需。 Objective To study the content of NO/MDA/SOD in 2,4,6-trinitrobenzene sulfonic acid(TNBS) induced ulcerative colitis(UC) in rats. Methods Total 38 SD rats were divided randomized into 5 groups as follows: normal group(5), one-day(6), three-day(7), seven-day(6), fourteen-day(6), twenty-one-day(7) groups, Besides the normal group rats, all of the other rats were induced by intrarectal administration of TNBS, and then observe the common conditions, colon length, ulcerative fraction and NO/MDA/SOD of the colon mucosa. Results Compared to the normal group, the colon was shortened and the ulcerative fraction was increased most obviously in the senven-day group( (P 〈 0.01) ), the others were all ( P〈 0.05) ; While NO content of the one-day, three-day groups in colon mucosa was increased, their significances being (P〈0.01), (P〈0.05) respectively; The MDA showed ascending trend in the oneday, three-day, seven-day groups, especially the three-day, seven-day groups were more significant(P〈 0.05) ;There was no significance in SOD between the normal group and the model groups(P 〉0.05). Conclusion The rat model of ulcerative colitis induced by TNBS could be content to the study of UC experiments.
