超声助溶治疗大鼠急性脑梗死 被引量:8

The therapeutic effects of low frequency ultrasound enhanced thrombolysis on acute cerebral infarction in rats

摘要 目的观察体外超声辅助尿激酶溶栓治疗急性脑梗死大鼠的效果。方法取84只Wistar大鼠,随机分为梗死组、大剂量尿激酶组(尿激酶组)、超声加小剂量尿激酶组(超声组)和假手术组。采用自身血栓栓塞法制备大鼠急性脑梗死模型,尿激酶组静脉给予尿激酶,剂量为1.5万U/只;超声组静脉给予剂量为1万U/只的尿激酶,同时经颅行超声治疗(频率为800 kHz,声强为1.2 W/cm^2,占空比为1:5)。采用NSS评价大鼠治疗前、后的神经功能,TTC染色测量脑梗死灶体积。结果尿激酶组和超声组治疗后24 h,NSS评分均明显低于治疗前(P<0.01)及梗死组(P<0.01),梗死灶体积也显著小于梗死组(P<0.01),但尿激酶组与超声组之间差异均无统计学意义(P>0.05)。结论体外超声助溶可促进急性脑梗死大鼠神经功能的恢复,减小脑梗死灶体积,减少尿激酶的用量。 Objective To observe the therapeutic effects of low frequency ultrasound enhanced thrombolysis (LFUET) on acute cerebral infarction (ACI) in rats. Methods The ACI animal models were established by injecting auto-thrombus into the rats' left middle cerebral arteries. They were then treated with urokinase, and received transcranial LFUET treatment at the same time. Nervous system functioning was assessed using NSS, and infarct volumes (IVs) were measured through tetrazolium chloride (TTC) staining. Results The NSS scores in the largedose urokinase group (LDU group) , the ultrasound plus small-dose urokinase group (USMU group) and in the infarct group ( I group) at 24 h after treatment were significantly lower than those before treatment. IVs in the two treatment groups are lower than those in the I group, but there was no significant difference between the LDU group and USMU group volumes. Conclusion LFUET can accelerate the recovery of nervous system function in rats after ACI, minimize IVs, and reduce the required dosage of urokinase.
