封闭负压引流技术对人慢性创面周围组织中PDGF及TGF-β表达的影响 被引量:40

摘要 目的研究人慢性创面用封闭负压引流治疗前后,创周组织中血小板衍化生长因子和转化生长因子-β表达的变化,探讨封闭负压引流技术加速创面愈合的机制。方法对6例慢性创面患者给封闭负压引流治疗,分别于治疗前及治疗后1、3、5、7d,对创周组织取材、匀浆、离心后得到创周组织的蛋白溶液,应用Western blot方法和Kodak Digital Science ID软件系统观察和分析PDGF及TGF-β的动态变化。结果治疗前创周组织中PDGF及TGF-β表达较少,治疗后两者表达逐渐增多,治疗后第3天起较治疗前显著增加。结论封闭负压引流技术可以提高慢性创面创周组织中PDGF及TGF-β的表达,从而促进创面愈合。 Objective To investigate the expression of platelet derived growth factor(PDGF) and transforming growth factor- 6(TCF-β) in margin tissue of human chronic wounds treated by vacuum assisted closure at different timepoints and explore the mechanism of accelerating healing of vacuum assisted closure. Methods Margin tissue of chronic wounds from patients were taken at 0, 1, 3, 5 and 7 days during the period of vacuum assisted closure treatment. The western blot method was applied to characterize the expression of PDGF and TGF - β in the above - metioned tissue and the results were analyzed by kodak digital science ID software system. Results The expression of PDGF and TGF - 6 in margin tissue increased significantly with the time of treatment prolonging. The expression of both expressed less at o day than those at 3, 5, 7 days. Conclusion VAC can increase the expression of PDGF and TGF - β, which is beneficial to the healing of chronic wounds.
