刺五加在心肌细胞缺血再灌注损伤中的保护作用 被引量:3

摘要 目的:研究刺五加注射液对缺血再灌注损伤(ischemia-reperfusion,I/R)心肌细胞的保护作用。方法:体外培养心肌细胞并计数;721可见光分光光度计测量培养液中的乳酸脱氢酶(LDH)、丙二醛(MDA)和超氧化物歧化酶(SOD)的含量;激光扫描共聚焦显微镜观察细胞内钙离子荧光染色。结果:中药刺五加注射液各剂量组与I/R组相比,细胞存活率升高,LDH、MDA水平降低,SOD水平升高,荧光像素值及阳性染色面积降低,有统计学意义。结论:中药刺五加注射液对缺血再灌注损伤心肌细胞有保护作用,可能与其保护细胞膜、清除氧自由基和减少钙离子内流等机制有关,其最佳浓度为40%。 Objective: To study the mechanism of the protection of acanthopanax on ischemia-reperfusion injury of myocardial cells. Methods: Myocardial cells were cultured in vitro and numbered. LDH, MDA and SOD in the culture fluid were measured with 721 visible range spectrophotometer and the fluorescence staining of the intracellular calcium ion were observed with division reproduction laser confocal microscopy. Results: Compared to the I/R group, the 3 acanthopanax groups had a high survival rate of myocardial cell, and SOD value, anti had a low LDH, MDA, fluorescence staining value and positive Ca^2+ staining areas. Conclusion: Acanthopanax has a pro tective effect on I/R isolated myocardial cells, the best concentration of which is 40%. It may be its mechanism to protect the cell envelope, clean oxygen free radical and decrease Ca^2+ inflow.
