摘要
Dab2/DOC-2是具有信号转导功能的磷蛋白,在丝裂原信号转导途径中起作用,通过蛋白质的相互作用和蛋白磷酸化发挥作用,参与控制细胞生长的信号通路而发挥生长抑制效应,Dab2/DOC-2又作为胞质衔接蛋白,参与细胞内蛋白运输。在肿瘤中纯合子基因缺失时被激活,是抑癌基因的候选成员。其在多种肿瘤中低或失表达,如卵巢癌,乳腺癌,绒毛膜癌和前列腺癌等,重新表达能抑制肿瘤的生长。Dab2也在胚胎的发生过程中发挥重要的作用。
DOC-2/DAB2 is a phosphoprotein having signal trnnsduction , which plays a role in mitogenic signal transduction pathway as well as growth inhibiting by controlling the signal pathway of cell growth through the interaction of protein and phosphorylation of protein. It also acts as cytoplasmic adaptor proteins psrticipating the intracellular protein transport, which can be activated by homozygote gene deletion in tumor and is a member of anti-on-cogene. It ofen has low or no expression in many tumors including ovarian cancer, breast carcenoma, chorionic careinoma, prostatic carcinoma and so on, whose expression renewing can inhibit the growth of tumor. Dab2 also plays an important role in the process of embryonic development.
