摘要
目的:建立水提三七叶苷副产物多糖的提取、分离及含量测定方法。方法:水提三七茎叶总皂苷废弃液经纳滤膜浓缩5倍后,低温加热浓缩至密度为1.25,乙醇多次沉淀提取多糖,用3,5-二硝基水杨酸(DNS)比色法以485nm为检测波长测定多糖含量。结果:浓缩液中多糖得率39.46%,含量为15.44%,方法的回收率为100.67%,RSD=2.74%。结论:从水提三七叶苷废弃液中提取多糖有较大开发利用价值;DNS比色法测定多糖含量操作简单,重现性好,可作为三七中多糖含量测定的常规方法。
Objective: To establish a feasible method for extracting, separating and determining polysaccharide from discarded liquor of stems and leaves of panax notogenseng after saponins has been extracted.Methods: The discarded liquor of stems and leaves of panax notoginseng after water extraction was condensed 5 times and hypothermally heated to the density of 1.25 through nano-filtration,from which the polysaccharide was obtained from alcohol precipitation.3,5-Dinitrosalicylic(DNS) colorimetry was used to determine the content of the polysaccharide by 485nm wavelength.Results: The rate of yielding the polysaccharide from the condensed liquor was 39.46%,the content was 15.44%,the rate of recovery 100.67% and RSD was 2.74%(n=6).Conclusion: The extraction of the polysaccharide from discarded liquor of stems and leaves of panax notogenseng has a great value of development and utilization.
