摘要
用人工合成的bcr3/abL2反义寡聚脱氧核苷酸(ASO-b)及无关寡聚脱氧核苷酸(N-ODN)处理人急性红白血病细胞株K562细胞,结果造成了细胞恶性行为的改变。表现为细胞生长减慢、P210蛋白表达受抑、DNA的合成减少及集落形成能力下降,从而证明了反义核酸在癌基因表达上的调控和抑制细胞恶性行为的作用。
The effects of antisense oligodeoxynucleotide on the expression of bcr3/abL2 proto oncogene and the growth of human cell line K562 were observed.Synthetic 18-mer ASO direct ed at the region of the translation initiation codon 2~7 of bcr3/abL2 proto-oncogene(ASO-b)greatly inhibited the proliferation of K562 cell line and the expression of P210 protein in human cell line K562.The synthesis of DNA and the ability of forming colony were also significantly suppressed.These results implicate the role of antisense oligodeoxynucleotide in the regulation of the expression of proto-oncogene and the reduction of maligant growth in cells.
