鼻咽癌自发性高淋巴道转移细胞亚群裸鼠体内筛选的研究

In Vivo Selection of a Cell Subpopulation of Nasopharyngeal Carcinomas with Spontaneous Highly Lymphatic Metastasis in Nude Mice

将低分化鼻咽癌细胞系（ＣＮＥ－２Ｚ）克隆株Ｈ５接种裸鼠皮下，待裸鼠发生转移后，取其部分淋巴结转移灶癌组织回复培养，并将回复培养的癌细胞再次接种裸鼠背部皮下，如此连续四代用淋巴结转移灶癌细胞接种传代，观察裸鼠自发性转移情况。结果显示：当淋巴结转移灶癌细胞连续接种传至第四代时，裸鼠淋巴结的转移率为１００％，肺转移率为９％。结果表明：鼻咽癌克隆株通过四代淋巴结转移灶癌细胞的体内筛选，已成为一鼻咽癌自发性高淋巴道转移的细胞亚群，这一细胞亚群的获得为鼻咽癌淋巴道转移机理的研究提供了极好的实验材料。

The subclones CNE-2Z-H5 of nasopharyngeal carcinomas(NPC)were inocubated into subcutaneous in nude mice.The parts of tissues of lymph node metastatic foci were cultured in vitro and than were inocubated into subcutancous in nude mice again when metastases of transplanted tumors were observed.After four selection cycles like above,the characteristics of spontaneous metastases was observed. The results showed that the lymph node metastatic rate was 100% and lung metastatic rate was 9% after four selection cycles in vivo.The results suggested that a cell subpopulation with sponotaneous highly lymphatic metastasis have been obtained after the subclones CNE2Z-H5 was selected by four passage cycles in vivo,and obtaining a good experimental materials for studing the mechanisms of lymphatic metastasis in NPC.
