摘要
目的研究Bcl-2、Survivin和CD44v6在宫颈癌组织中的表达及其与细胞凋亡、HPV16/18感染的相关性,以探讨宫颈癌的发生机制,指导其诊断预后。方法采用免疫组化对宫颈组织石蜡标本进行Bcl-2、Survivin、CD44v6的检测,同时采用TUNEL和PCR技术检测细胞凋亡指数(AI)和HPV16/18感染情况。结果Bcl-2、Survivin、CD44v6的阳性率和AI在宫颈癌组织中与CIN、正常宫颈组织有差异,且随着临床分期、病理分级的升高阳性率有差异,其表达率与淋巴结转移有关。HPV16/18在正常宫颈组织、CIN和宫颈癌中的阳性率逐渐升高,差异有统计学意义。Bcl-2、Survivin和AI均与HPV16/18感染有相关性。结论宫颈癌组织中Bcl-2、Survivin、CD44v6的异常表达和HPV16/18感染有关,与宫颈癌的恶变程度有关,可作为宫颈癌筛查预后判断的有利指标。
Objective To investigate the relationship between expression of Bcl-2, Survivin and CD44v6 and HPV16/18 infection in uterine cervical carcinogenesis. Methods Formalin-fixed, paraffin-embedded samples from cervical carcer tissues were studied, the authors examined the expression of Bcl-2, Survivin and CD44v6 by using immunohistohemical technique. The number of apoptotic cells was assessed in situ by TUNEL method. The infection of HPV type 16,18 DNA were determined by PCR. Results There were significant differences for Bcl-2, Survivin, CD44v6 and AI between carcinomas, CIN and normal cervices, respectively. Expression of Bcl-2, Survivin, CD44v6 and AI were correlated with tumor grades, clinical stages and lymph node metastasis. The positive rate of HPV16/18 was significantly different among cervical carcinomas, CIN and normal tissues. The infection of HPV16/18 was associated with the expression of Bcl-2, Survivin and AI, respectively. Conclusion The abnormal expression of Bcl-2, Survivin and CD44v6 are associated with the infection of HPV16/18. They possibly become useful indexes for the primary screening and prognosis of cervical carcinomas.
