2种大豆总RNA提取方法的改良 被引量:11

Two efficient improved methods for isolation of high quality total RNA from soybean

摘要 对大豆总RNA的提取方法T rizo l法和异硫氰酸胍法进行了改良,比较了改良前后2种方法的提取效果。结果表明,2种改良方法提取的大豆总RNA质量较高、完整性较好,完全可以满足后续研究要求,RNA产率均超过220μg/g。改良T rizo l法适用于大豆根、茎、叶及生长点总RNA的提取,而改良异硫氰酸胍法适用于大豆根、茎及生长点总RNA的提取。 Two methods of isolating RNA were improved in this paper. The effects of extracting total soybean RNA with the improved methods were compared with those original ones. The result shows the improved Trizol method fits for extracting RNA from soybean root ,stem,leaf and shoot apical mer,whereas the improved Guanidinium Thiocyanate method fits for root,stem and shoot apical meristem (SAM). More than 22 μg RNA was obtained from each 100 mg plant tissue with high purity and integrity. Therefore ,these methods deserve further studies.
