摘要
目的利用白花蛇舌草茎尖的愈伤组织建立细胞悬浮培养系,并通过实验研究不同条件对白花蛇舌草细胞悬浮培养细胞生长的影响。方法采用单因素试验对不同种类、不同浓度的激素及其组合、碳源的种类以及碳源浓度、起始的pH值对白花蛇舌草细胞悬浮培养细胞生长的影响作用进行研究。结果最适pH=6.5,激素以NAA效果最好,其中NAA最佳浓度为1.5mg·L-1。组合激素对细胞的生长的促进作用和单种激素相比,没有明显增强。蔗糖比葡萄糖更适合作为碳源,蔗糖的最佳浓度为30g·L-1。结论本试验建立了白花蛇舌草的细胞悬浮培养系,提出了培养的最优条件,可以用以大量生产。
OBJECTIVE To build Hedyotis Diffusa cells suspended culture with the callus of the stem's tip, and observe the effect on the conditions to it. METHODS The sigle element tests was used to study the effect about various conditions, such as hormones and hormone combination, the type and concentration of sugar, the primary pH. RESULTS Optimal conditions is pH = 6.5, the most suitable hormone was NAA. and its concentration was 1.5mg ·L^-1, hormone combination'effect is not stronger than single hormone. Saccharose is more suitable than glucose,and its concentration was 30g · L ^-1.
