丙酸菌生长细胞转化亚油酸生成共轭亚油酸 被引量:5

摘要 本文对费氏丙酸杆菌HZ—P-35转化亚油酸生成共轭亚油酸基本特性进行了研究。共轭亚油酸的测定以十七碳烯酸作内标物经甲酯化后采用气相色谱法测定。实验结果表明,费氏丙酸杆菌菌株HZ—P-35在改良MRS培养基中厌氧培养时生长较为缓慢,96h左右对数生长期结束;接种量10%有利于菌株的生长及共轭亚油酸转化;最适温度30℃既有利于菌体生长又有利于共轭亚油酸的生成;细胞边生长边转化底物亚油酸,培养4~5d时共轭亚油酸转化迭最大值,而至第6d菌体浓度具有较大值。 Propionibacterium frudenreichii strain HZ -P -35, isolated from cow feed, was studied for its conjugated linoleie acid (CLA)production from linoleic acid. CLA determination was conducted by GC. The following are the results : Strain HZ - P - 35 has a slow growth in a modified MRS medium with a 96h time course of logarithmic growth phase. This strain can grow between 25 -40%, but the, optimum temperature is 30% while cell growth and CLA conversion performance both better. Inoculation volume of 10% (v/v) is proper for growth and conversion yield. After 4 -5 days cell growth and conversion, CLA concentration is up to 109.8μg/mL, coincided with the quick cell growth. The maximum of biomass concentration can be obtained at 6th day.
