摘要
目的研究缬沙坦对阿霉素(ADR)致心肌损伤大鼠心功能及心肌肌球蛋白重链(MHC)基因表达改变的相关分析。方法30只SD大鼠腹腔注射ADR(20mg/kg,分8次)造成心脏损伤模型,缬沙坦330mg/kg灌胃,分11次观察其对ADR致心肌损伤的保护作用。停药24h后,测定大鼠左心室功能,肌球蛋白重链分析(半定量)。结果①α-MHCmRNA基因表达减弱,β-MHCmRNA基因表达增强;②心肌α-MHC向β-MHC的表型转变与心功能的损伤具有显著相关性。结论缬沙坦能逆转α-MHC向β-MHC的表型转变,心室功能的改善与MHC表型改变的逆转具有显著相关性。
Objective To investigate the relationship between the protective effects of valsartan on myocardium and the expression changes of cardiac myosin heavy chain(MHC) genes in rat models of experimental heart injury induced by adriamycin(ADR), Methods Thirty SD rat models of experimental myocardial injury were established by eight times of intraperitoneal injection of ADR with a total dosage of 20 mg/kg. Valsartan(330 mg/kg) was given through gastric clysis. Twenty-four hours after the clysis, the function of the left heart was detected and the expression of the cardiac MHC genes was analyzed, Results The expression of α-MHC mRNA was decreased and the expression of β-MHC mRNA was increased in ADR group. The shifts of myosin from α-MHC mRNA to β-MHC mRNA was correlated with heart injury. Conclusions Valsartan could play a role in the shift from α-MHC mRNA to β-MHC mRNA. The improvement of heart function could reverse the shift of the expression of MHC genes.
