摘要
[目的]从棒曲霉菌培养上清液中分离纯化具有抗真菌活性的肽类物质(AFP).[方法]用离子交换柱层析法分离AFP,利用反相高效液相色谱法进一步纯化,经聚丙烯酰胺凝胶电泳分析确定相对分子质量.[结果]从棒曲霉菌培养上清液中分离出1种具有抗真菌活性的AFP,纯化后得到高活性的精制AFP,相对分子质量约为8 ku,MTT法测定结果表明AFP对5种真菌均具有明显的抗真菌作用.[结论]棒曲霉菌培养上清液中含具有抗真菌活性的AFP.
OBJECTIVE To purify the peptide with antifungal activity from Aspergillus clavalus desm culture fluid. METHODS The peptide was purified by reverse phase HPLC, and the separa molecul ted by ion exchange column chromatography and further ar weight was determined by tricine gel electrophoresis RESULTS A kind of peptide with antifungal activity was separated. The molecular weight was about 8 ku. The detection result by MTT method showed significant antifungal activity of the peptide to 5 kinds of fungi. CONCLUSION There has antifungal peptide in Aspergillus clavalus desm culture fluid.
