摘要
用荧光光谱法研究了人血清白蛋白与槲皮素的结合反应,发现槲皮素对人血清白蛋白的荧光有很强的猝灭作用,求得它同人血清白蛋白的离解常数(k_D=5.43×10^(-5)。鉴于人血清白蛋白与槲皮素之间可发生能量转移,利用荧光共振能量转移原理测定人血清白蛋白的色氨酸与槲皮素之间的距离,提出了复合物结合模型并探讨了其结合机理。
The binding of quercetin to human serum albumin was studied using fluorescence spectrum. It was shown that it had powerful ability to quench the human serum albumin fluorescence via a ninradiative energy transfer mechanism. The dissociation constant (kD:5. 43×10-5) was obtained according to analyzing the fluorescence quenching date. By use of a spectra overlap integral between the absorption spectrum of it and emission spectrum of human serum albumin, the distance of human serum albumin to quercetin was estimated. More over, the binding mechanism was discussed in brief and the quercetin-human serum albumin complex was postulated from above results.
