2型糖尿病大鼠胰岛11β-类固醇脱氢酶1型的表达对β细胞功能的影响 被引量:3

Influence of islet 11β-HSD1 expression on isletβ-cell function in streptozotocin and high-fat induced diabetic rats

摘要 建立链脲佐菌素联合高脂喂养诱导的2型糖尿病(DM)大鼠模型并进行腹腔内葡萄糖耐量试验,评价β细胞功能,RT-PCR和Western印迹结果显示2型DM模型大鼠胰岛11β-类固醇脱氢酶1型(11β-HSD1)表达明显升高,与空腹血糖水平呈正相关;与空腹胰岛素、胰岛素和血糖曲线下面积比值呈负相关;且11β-HSD1表达与Pdx1表达负相关。提示2型DM模型大鼠11β-HSD1在胰岛表达增加,扩大局部组织糖皮质激素效应,使B细胞功能受损。 Intraperitoneal glucose tolerance test was performed in the streptozotocin- and high fat-induced diabetic rats and normal rats. The results of RT-PCR and Western blot showed that the expression of 11β- hydroxysleroid dehydrogenase type 1 ( 11 β-HSD1 ) was higher in the diabetic rats than that in control and was correlated with fasting plasma glucose, insulin and AUC-I/G with respective correlation coefficient (r) of 0. 870, - 0.799, - 0. 850, suggesting that increased expression of 11 β-HSD1 appears to damage β-cell function through magnifying the local effect of glucocorticoids.
