摘要
在单波长紫外可见分光光度计上测定样品在不同波长(λ)下的吸光度(A),然后利用数学处理方法将-λA转变为不同波长下的一阶、二阶及三阶导数关系,即λ-ΔA/Δλ、λ-Δ2A/Δ2λ及λ-Δ3A/Δ3λ关系,并在计算机上利用EXCEL绘制出样品的各阶导数吸收光谱。实现了在单波长紫外可见分光光度计上测定导数吸收光谱的目的,扩大了单波长紫外可见分光光度计的应用范围。
Determination of derivative absorption spectra with conventional or monowavelength UV-VIS spectrophotometer was performed by measuring the absorbances (A) of the sample at different wavelength (λ), and then converting the serial data of A and λ to values of 1st, 2nd and 3rd derivative at different wavelength, i.e. the relationships between λ-△A/△λ、λ-△^2A/△λ^2,and λ-△^3/△λ^3 by mathematical calculations, and plotting the respective derivative absorption spectra on a computer with the software EXCEL. By the use of mathematical method and computer technique in combination with spectrophotometry, the function of conventional UV-VIS spectrophotometer was extended to derivative spectrophotometry.
