摘要 研究了在酸性溶液中偶氮氯膦Ⅰ(CPA-Ⅰ)-铁(Ⅲ)配合物与牛血清白蛋白(BSA)结合的吸收光谱行为,建立起一种以偶氮氯膦Ⅰ-铁(Ⅲ)配合物作为光谱探针测定蛋白质的新方法。该法具有较强的抗干扰力,弥补了偶氮氯膦Ⅰ测定蛋白质干扰物质允许量低的问题,该法反应灵敏、快速,摩尔吸光系数5ε70nm=2.70×105L.m o-l 1.cm-1,线性范围宽,可达0—82m g/L,用于人血清总蛋白的测定与双缩脲法相关性好。 A new method for the determination of protein by using chlorophosphonazo Ⅰ (CPA- Ⅰ )-ferrum ( Ⅲ ) complex as a spectral probe was established based on the binding interaction of protein with CPA- Ⅰ -ferrum ( Ⅲ ) complex in acid buffer. The method shows high selectivity, and overcomes the problem of interference in the determination of serum proteins by using CPA- Ⅰ . This method is sensitive and rapid. The molar absorption coefficient is 2.70 × 10^5L·mol^-1·cm^-1. The calibration graph for BSA is linear up to 82mg/L. The method has been employed in the assay of protein of human serum. The results are in agreement with those obtained by Biuret method.
