测定花生及花生制品中黄曲霉毒素:免疫亲和柱净化·光化学在线衍生·高效液相色谱-荧光 被引量:6

Detecting Aflatoxin in Peanut and its Products by Immunoaffinity Column,Photochemical Derivatization and High Performance Liquid

摘要 〔目的〕建立免疫亲和柱净化、柱后光化学在线衍生、高效液相色谱-荧光法测定花生及花生制品中黄曲霉毒素B1、B2、G1和G2的检测方法。〔方法〕样品粉碎、由甲醇/水提取、Afla-P黄曲霉毒素免疫亲和柱净化、光化学衍生器在线衍生、高效液相色谱-荧光法检测。〔结果〕黄曲霉毒素B1、B2、G1和G2的检出限分别为0.5、0.15、0.5、0.15μg/kg,相对标准偏差低于15%,回收率为64%~97%。〔结论〕该方法简便快速、灵敏准确、重现性好,能够满足出入境检验检疫工作的需要。 Objective To set up the method of Afle-P immunoaffinity colum,photochemical derivatization,and high performance liquid chromatography. Method Sample of Peanut and its products were extracted with MeOH-H2O (6+4,v/v). Extract was filtered, diluted with same volumn of water, and applied to Afla-P affinity column containing antibodies specific to aflatoxins B1,B2,G1 and G2. Aflatoxins were removed from the affinity column with MeOH and quantified by reversed-phase chromatography with post-column photochemical derivation and fluorescence detection.Result The limit of determination was 0.5,0.15,0.5,0.15μg/kg,the relative standard deviations was below 15%.Recoveries of Aflatoxin B1,B2,G1 and G2 spiked to samples were between 64%-97%.Conclusion This method is oimple and conveniecnt, rapid,sensitive,and exact.It would meet the need of entry-exit inspection and quaratine work.
